I often see the Boeing RC-135 but the have all time an other Callsign. 
Does anyone know why?

Callsigns will change as they do for an airline's flights.

My guess a new callsign is allocated for each sortie.

I don't think that's correct.  Not all "US Mil Flight" use the same callsign.  Not the ones I see.

for example i see the callsign "Lion" with random number.

For what it is worth call sign "Lion" is a long standing Belgium Military assignment.
